Any particular reason? I mean, aside from Legendaries and Starters, every Pokemon that has been in any game is catchable in Diamond/Pearl*.

Mewtwo can be obtained in Fire Red/Leaf Green. Transferable to D/P via the Pal Park.
Mew is only available by going to Pokemon events like at Toys R Us. There was a Mew one not too long ago. I got one, and it shall probably be in my party as soon as I have access to the Pal Park.

If you own LeafGreen/FireRed, you can transfer a Mewtwo from one of those games into Diamond/Pearl once you unlock Pal Park, which requires that you see all 150 of the Pokemon in D/P's Pokedex.
Mew can be captured in Emerald, but you'll need to have gone to a Nintendo event to get the item that unlocks it. Easiest way to get it, IMO, is to have a Game Shark/Action Replay, and hack in the item. Once you've caught the critter, you can bring it into D/P via the same method as Mewtwo.
