Tip 1
The Windows+F searchbox actually sucks a lot for general searching.
It's slow, and if the index is incomplete it will return false negatives. It's only real use is doing large-scale, full-system queries, to get a systemwide list of files sorted by date modified, or by size for example.
Better for general searching is to do the following. In explorer, to go folder options
go to the search tab and configure it like this
Then use the integrated search box from whatever directory you'd like to search.
It'll give you results in a view customized to fit the results. My C:\stuff\ folder is set up to display in Details view by default, but because I searched for Tits and most of the results were images, Vista automatically chose a thumbnail view.
Many of Vista's functions seem tailor-made for pornography browsing.
Tip 2
When you click the Windows Orb of Power and open the menu formerly known as start, you'll notice another fun little search box.
This guy also brings you the POWER OF THE COMMAND PROMPT. All your favorite Win+R run box tricks also work directly from this search box.
Tip 3
Hold shift and right click on a file for HIDDEN CONTEXT MENU OPTIONS
The options that show up vary between file types, but those 3 are always there. Copy As Path is especially handy imo.

This would be way more useful if the command prompt window stayed open after the command had completed. As it is, I find myself fumbling with syntax often enough that I need to open a command prompt anyway. `shutdown`'s options are particularly hard to remember, for some reason.
I also really wish that text fields had useful keybindings; in OS X, all Cocoa text fields use a subset of emacs keybindings. There's a whole class of commands which I just have no idea how to do under Windows, like jump around by word, delete word, kill line, etc.
